Describes new methodology for reducing time spent single-cell cloning a cell population with a knockout deletion.
The authors developed two methods to select nuclease-edited cells. Co-transposition (now known as GEEK) and co-targeting utilize induced phenotypic changes that allow for enrichment of gene edited cells. Unlike FACS and reporter-based enrichment strategies, the methods presented are simple, efficient, allow for isolation of single-cell derived clones. These methods expedite the generation of knockout cells lines and have the potential to improve the generation of therapeutic cells.
